I have a 2007 Highlander with the 3.3L V6 engine. Does this engine have a
cast iron or aluminum block.

I cannot tell by what I can see. Could be lots of silver paint on iron.

I don't know, why don't you try sticking a small magnet to the
block and see if it sticks?
